LABEL POSITION MATTERS

Custom isotope-labeled lipids

Put the label where the question is.

When the available labeled analogue does not match your analyte or tracing question, a custom labeling pattern may be the better starting point. Headgroup-labeled variants are a particular focus; other sites can be discussed.

RESEARCH-SPECIFIC, BY DESIGN

Example custom requests.

Illustrative starting briefs. Each requires confirmation of the exact structure and feasibility.

01

A headgroup-labeled analogue

A requested PC or PE structure with a defined headgroup labeling pattern, rather than a label in the fatty-acid chains.

02

A missing internal-standard candidate

A labeled counterpart of a hard-to-source lipid for evaluation in a targeted analytical method.

03

A position-specific tracer

A lipid with a specified label location to investigate the part of the molecule relevant to a research pathway.

details clear.

Name the atoms, not just the mass shift.

Specify the isotope, number of labels and their positions when known. Include the desired enrichment and whether a distribution of isotopologues is acceptable. A drawing marking the atoms is especially useful.

Describe how the label will be used.

For a tracing project, explain which part of the molecule you intend to follow. For an internal standard, identify the target analyte, analytical platform and relevant ions. The labeling plan should support that use rather than simply create a heavier molecule.

Define the analytical acceptance criteria.

Chemical purity and isotopic enrichment are separate specifications. Agree on the evidence required for each, the amount needed and the presentation. A custom compound still needs validation in the researcher’s own analytical method; its availability does not establish method performance.

BEFORE YOU START

A few useful answers.

Are labels restricted to the fatty-acid chains?

No. Headgroup labeling is a focus, and other defined labeling positions can be discussed. Feasibility depends on the exact molecule, label pattern and available starting materials.

Is any isotope-labeled lipid a suitable quantitative standard?

Suitability depends on the analyte and method. Specify the intended measurement and validation requirements when requesting the material.
